This significantly outperforms the theoretical 13.5% yield for a 100 nt sgRNA, which demands a challenging-to-maintain 98% stepwise coupling efficiency for long synthesis and is restricted to gram-scale deliveries in industry. N-1 impurities in a 32mer are easily removed by preparative HPLC or denaturing PAGE, unlike sgRNA, where N-1 impurities remain persistently difficult to decrease. All lgRNA segments are shorter than 36 nt, ensuring compatibility with industrial oligonucleotide manufacturing at kilogram batch scales.
